Abstract

Background: Chlorhexidine is a broad-spectrum antimicrobial agent widely used for the treatment and prevention of oral diseases. However, the therapeutic efficacy of conventional dosage forms is limited by their short residence time at the site of action, particularly in the local treatment of periodontal disease. Oral films have emerged as a promising drug delivery system capable of prolonging drug retention and providing controlled drug release.

Objectives: This study aims to develop the formulation and manufacturing process of chlorhexidine-loaded oral films and to evaluate their physicochemical characteristics.

Materials and methods: Chlorhexidine, hydroxypropyl cellulose (HPC), hydroxyethyl cellulose (HEC), polyethylene glycol 400 (PEG 400), and other excipients were used. The films were prepared by the solvent casting method. Formulation composition and processing parameters were investigated based on film appearance, folding endurance, and swelling behavior. The selected formulation was further evaluated for its in vitro drug release profile.

Results: The selected formulation consisted of 1.5% HPC, 1.5% HEC, and 0.5% PEG 400 dispersed in distilled water, with 5 g of casting gel per mold. The resulting films contained 5.65 ± 0.21% (w/w) chlorhexidine and exhibited a thickness of 0.23 ± 0.01 mm, a folding endurance greater than 300, and a surface pH of 5.85 ± 0.30. The films released 72.37 ± 8.78% of chlorhexidine after 6 hours of in vitro dissolution testing. The in vitro release kinetic was best explained by Higuchi model.

Conclusions: A chlorhexidine-loaded film formulation and preparation process were developed, showing potential for further development as a drug delivery system for the treatment of periodontal disease.
