Abstract
Background: Holarrhena pubescens has been widely used in traditional medicine and various folk remedies. However, studies on the quantification of conessin-the main active compound-and the biological activities of this medicinal plant remain limited. This study aims to develop a quantification method for conessin using high-performance liquid chromatography (HPLC) and evaluate the in vitro anticancer and anti-inflammatory activities of the stem bark extract of Holarrhena pubescens.
Materials: Stem bark of Holarrhena pubescens.
Results: The quantitative method demonstrated high specificity, suitability, linearity, precision, and accuracy. The extract exhibited cytotoxic activity against the SK-LU-1, HepG2, and SW480 cancer cell lines, with IC₅₀ values ranging from 45.55 to 67.70 µg/mL. Additionally, it demonstrated NO production inhibition, with an IC₅₀ value of 90.83 µg/mL.
Conclusions: The developed HPLC method is applicable for the quantitative determination of conessin in Holarrhena pubescens samples. The stem bark extract exhibited significant in vitro anticancer activity against three tested cell lines and also showed anti-inflammatory potential. Notably, this study is the first to report the in vitro anticancer effects of the stem bark extract of Holarrhena pubescens on SK-LU-1, HepG2, and SW480 cell lines.
