Abstract
Background: The IL10 gene -819T>C (rs1800871) polymorphism has been associated with various diseases, including miscarriage, cancer, stroke, and osteoporosis... This study aimed to: (1) Develop an allele-specific PCR assay using a self-designed primer pair to identify the IL10 gene -819T>C polymorphism (rs1800871), and (2) Investigate the genotype and allele distribution of IL10 gene rs1800871 polymorphism in healthy volunteers. Materials and Methods: Based on the IL10 gene sequence containing the rs1800871 polymorphism, two forward primers was designed with the 3’-terminal nucleotides corresponding to either the T or C allele of the polymorphism. The reverse primer was designed using PrimerDigital, an online software. Positive control samples were identified using Sanger sequencing. PCR settings were optimized and applied to identify the IL10 rs1800871 polymorphism in 100 healthy volunteers including 67 women and 33 men. Results: An allele-specific PCR procedure was successfully developed to identify the IL10 -819T>C (rs1800871) polymorphism. The genotype frequencies of -819TT, -819TC, and -819CC were 52%, 42%, and 6%, respectively. The -819T and -819C alleles had frequencies of 73% and 27%, respectively. Conclusion: We developed successfully an allele-specific PCR approach for detecting the IL10 -819T>C polymorphism. We determined the genotype and allele frequencies of the -819T>C polymorphism in healthy volunteers.
