Abstract
Background: Supramolecular deep eutectic solvents (SUPRADES) have recently emerged as promising green solvents for the extraction of bioactive compounds from medicinal plants. Objectives: This study aims to develop a green extraction strategy for isolating rutin and quercetin from Sophora japonica using SUPRADES. Materials and method: Various SUPRADES systems were screened and evaluated for their extraction performance. The extraction conditions were optimized using response surface methodology. The antioxidant activities of the resulting extracts were assessed and compared. The greenness of the proposed method was quantitatively evaluated using the AGREEprep metric. Results: A SUPRADES composed of choline chloride/acetic acid and β-cyclodextrin was identified as the most effective and environmentally friendly solvent. Under optimal conditions, the total extraction yield of rutin and quercetin reached 35.21%, which was 2.16 times higher than that obtained with methanol. The antioxidant capacity of the SUPRADES extract was 2.11 times greater than that of the methanolic extract. The greenness score of the proposed method reached 0.79, indicating excellent environmental performance. Conclusion: These findings contribute to the development of a green extraction approach for flavonoids from Sophora japonica, and demonstrate the potential of SUPRADES as a safe, efficient, and eco-friendly alternative to conventional organic solvents.
